The Hippocampal Subfields Group hold conferences one or more times a year. This includes both smaller working group meetings for developing aspects of the harmonized protocol, as well as larger meetings designed to disseminate and discuss progress made by these working groups. The broader goal of these HSG meetings is to gather together a group of scientists interested in harmonizing across protocols and coming up with a unified subfield segmentation protocol. HSG meetings are open to HSG members and non-members alike.

In the lead-up to SfN, the hippocampal head working group convened over two days to advance the development of the hippocampal head segmentation protocol. In addition to the official working group members Marshall Dalton and Renaud La Joie, the meeting welcomed the participation of HSG members Kim Nguyen, Ana Daugherty, Kelsey Canada, Carl Hodgetts, and Rosanna Olsen.

The MTL working group gathered in Lund to further their efforts in developing a unified MTL protocol. Additionally, a Marcus Wallenberg Symposium in the form of an online webinar was conducted to delve into the recent advancements made by the HSG working groups.

This June, we held a HSG meeting in Albacete, Spain. The meeting consisted of a two-day neuroanatomy workshop, including a hands-on brain dissection session, and a two-day working group meeting continuing our efforts for both the hippocampal head and MTL cortex. This was a two-part meeting, the first of which took place at UCSD and was a smaller meeting of the Boundary Working Group. The second day of the meeting was open to the broader HSG and any interested attendees of SfN.

Broadly, the aims of this meeting were to:

  • Provide an update on the harmonization effort from the Boundary Working Group
  • Engage the broader community about current and future directions
  • Share research findings during an afternoon research symposium

Thanks to all who attended this productive meeting! It was extremely encouraging to learn about the progress made by members of the Boundary Working Group (BWG). Specifically, the BWG provided updates regarding development of a protocol to define: the outer boundaries of the hippocampal body (anterior/posterior, medial/lateral, inferior/superior), the inner boundaries of the hippocampal body (subiculum, CA 1, 2, 3, and dentate gyrus), and preliminary efforts to define subfields within the hippocampal head. We also learned about efforts from the Questionnaire Working Group to put these protocols into writing, which will first be disseminated to the BWG and then to the broader subfield community for feedback. We had a productive group discussion regarding the progress of these groups, and also heard an exciting series of research talks in the afternoon.

We would like to thank all the participants of this exciting and successful meeting. This intense two-day retreat was a major step forward: we succeeded at developing a first draft of an harmonized protocol for subfield segmentation of the hippocampal body.

The meeting started with presentations of the recent progress from multiple working groups.

- Outer boundary group (presentation by David Berron). How should we use and/or adapt existing harmonized protocols for hippocampal volumetry (see the EADC-ADNI Harmonized Protocol project website) to our specific high-resolution images? How should we handle white matter? CSF pools? How should we deal with the specific issues related to partial voluming? Where is the most medial boundary of the hippocampus/subiculum?

- Histology working group. Laura Wisse presented preliminary data on the behalf of the histology working group (J. Augustinack, R. Insausti, K. Amunts and O. Kedo), showing segmentation of hippocampal subfields on high-resolution histology slices from three expert groups. The goal of the group was to provide 'gold standard' histology segmentations of the hippocampal body from multiple individuals.

- Body ranging rules. Where is the hippocampal body? On which slice does it starts? Where does it end? Ana Daugherty presented preliminary data on the reliability of previously-determined landmarks. For that purpose, 9 volunteers had assessed the boundaries of the hippocampal body on high-resolution MR images nicely provided by several groups of the harmonization project.

Following these presentations, we constituted small working groups to discuss and make decisions about specific boundaries: i) CA1-Subiculum, ii) CA1-2-3 and, iii) CA3-DG. After multiple discussions, decisions were made regarding each of these boundaries.

Next steps will be necessary to carefully check these rules; for example to check them more thoroughly with histology and to discuss their generalization across individuals, ie. considering hippocampi with variable (flat, round) shapes. Then, we will initiate a small-scale reliability attempt with expert raters.
